Not sure about D-Tools, but I think it should be possible. Otherwise it's just to burn the CD32 game onto a CD-R and have this in the drive when you launch the WinUAE emulation.
But what that lets you emulate CD32, is simply by feeding WinUAE with the CD32 kickstart ROM files in the appropriate fields on the "ROM" page. So the key of emulating CD32 is practically only in the use of kickstart files.
You'll need a CD32 kickstart 3.1 ROM (not sure if you can use the same as the regular A1200 3.1 ROM though, or if it got its own) to insert in the "ROM File" field and a CD32 Extended kickstart ROM to be inserted in the "Extended ROM" field.
Then just configure WinUAE according to a CD32:
RAM: 2MB Chip
CPU: 68EC020
Chipset: AGA
You should then be able to emulate the CD32 and its games. Even get that neat colored startup-logo and the CD player (but of course, since that's in the kickstart ROMs)
