Hey, has anybody ever tried applying various colors using Copic Markers?

I use acrylics and am also wondering if they can be over sprayed with acrylics or applied over acrylics and then sealed

I would hesitate to use them.

Google-based research indicates they are alcohol-based. Reports indicate that Sharpie markers, also alcohol-based, will bleed through any paint applied over them. Tamiya makes some paint markers; I've used them without any problems.
