I made a quick check on this. All I can say is that Karateka seems to be using a disk turbo that is not supported by any of the emulators for now.
My emulator, Yape, has just got processor level drive emulation, which is essential to get a disk turbo to work. But not enough I still have timing problems, that I hope to sort out in the not so distant future. Only then Karateka might be running...
