To answer your questions. Yes, any class may be chosen as your first class. Depending upon your playing style: trap assassin or martial arts assassin, you might want to choose pyro, electro, or body. See my other reply for further info.
To clarify a few things about skill points in skills other than your chosen class. For example: your first class is Electrokinesis. In order to learn claws of thunder, you first need to learn fists of fire. That is fine. However, you cannot use fists of fire until you have completed the Izual quest in Act 4, and chose pyrokinesis as your second class. Understand? If a skill is needed to be learned because it is a prerequisit, that is fine as long as it is not used until the class it belongs to is selected.
To explain the free class and the passive skills. Yes, claw mastery and weapon block maybe learned as a free class skill after you have selected a class and already devoted a point into the chosen class. All other free class skills must wait to be used until after you have completed the Radament quest in Act 2.
